Synthesis of Silver Nanoparticles using Mukiamaderaspatana leaf extract and evaluation of its antioxidant, antidiabetic, dye reducing and anticoagulant properties –An in vitro approach

Abstract: Plant-based nanoparticles have attracted more attention due to growing interest in environmentally conscious products.Synthesising silver nanoparticles (Ag-NPs) through green synthesis results in less toxicity which favours its use for various ailments. Mukiamaderaspatana,is a well known folkloric traditional medicine of India. In the present study, an attempt has been made to synthesize nanoparticles using aqueous leaf extract of M.maderasapatana and evaluate its antioxidant and antidiabetic properties.The synthesized silver nanoparticles using M.maderasapatana were characterized by spectral studies. The biosynthesized Ag-NPs of M.maderasapatana showed total antioxidant activity of 416µg AAE/mg.The IC50 value of DPPH scavenging activity of the synthesized Ag-NPs was found to be 1mg/ml . The antidiabetic activity of Ag-NPs was shown by the effective inhibition against carbohydrate digestive enzyme, alpha amylase with IC50 value of 500µg/ml.The Ag-NPs prevented formation of blood clot when used as anticoagulation agent. Microscopic examination depicted dispersed RBCs in EDTA treated blood and Ag-NPs treated blood at a concentration of 1mg/ml. The biosynthesized NPs degraded malachite green and methylene blue dyes in the range of 62.5% and 73.3% respectively after 24hrs incubation. From the present study, it is inferred that Ag-NPs synthesized from M.maderasapatana leaf extract has significant antioxidant and antidiabetic potential which may be useful in management of post-prandial hyperglycemia and type-2 diabetes.
