Solubility and Dissolution Enhancement of Atorvastatin by Nanocrystal Approach

Abstract:
|
Atrovastatin a antihpertensive drug HMG-CoA inhibitor used in hyperlipidemia, exhibit poor aqueous solubity, consequently dissolution and bioavailabity. In the present investigation attempt has been made to improve the solubility and dissolution rate of atorvastatin by preparing its nanocrystal formulation by anti solvent precipitation technique using poloxamer 188. The process was optimized interms of concentration of stabilizer poloxamer 188 (0.125%, 0.25%, 1%, 1.5%, 2%) in order to obtain stable particles with small uniform size. The prepared nanocrystals characterized by SEM, FTIR, DSC, XRD and particle size. The solubility profile was compared with pure atorvastain and found that there is more than ten fold increase in solubility, also dissolution of the nanocrystal formulation shown to be improved as compare to pure drug.
