Novel nano-metal complexes of modified aspirin as future antimicrobial agents

Abstract: Aspirin which was initially developed as an analgesic anti-inflammatory agent has come to be the basis of antiplatelet therapy. It has been reported that, 10-60% of patients on aspirin therapy are resistant to aspirin and such patients are at risk of developing cardio-vascular events, therefore, we tried to modify its structure to be useful for multiple therapeutic effect. New Mn(II), Ni(II), Cu(II), Zn(II) and Cd(II) organo-metallic compounds in nanoform of modified aspirin have been prepared and characterized using elemental and spectral analyses such as IR, UV-VIS, 1H-NMR, mass spectra, ESR as well as conductivity, magnetism, thermal analysis (DTA and TGA) and electron microscope. Elemental and spectral analysis showed, all the complexes were octahedral structure, however, the conductivity measurement confirmed non-electrolytic nature except Cu(II) complex (4). The ESR spectra of solid state at room temperature for Cu(II) complexes (2), (4), (7) had an axial type (dx2-y2) with covalent bond character, Mn(II) complex (14) showed isotropic type with octahedral structure. In vitro antimicrobial screening test for the ligand and some of its complexes showed, for (+ve) (Staphylococcus aureus), the order is ligand (1)> Cd(II) complex (10)> ampicillin> Zn(II) complex (8)> Cu(II) complex (2) = Cu(II) complex (3)> Cu(II) complex (4) = Cu(II) complex (6) = Ni(II) complex (12) > Aspirin > Mn(II) complex (14) and for (-ve) (Escherichia coli), the order is ligand (1)> Cd(II) complex (10)> ampicillin> Zn(II) complex (8)> Cu(II) complex (3)> Cu(II) complex (4)= Cu(II) complex (6)= Ni(II) complex (12)> Cu(II) complex (2)= aspirin> Mn(II) complex (14) and (Aspergillus flavus), the order is Cd(II) complex (10)>amphotericin B> ligand (1) > Cu(II) complex (2) = Cu(II) complex (3) = Cu(II) complex (4) = Cu(II) complex (6) = Zn(II) complex (8)= Ni(II) complex (12)= Mn(II) complex (14) and (candida albicans), the order is amphotericin B > Cd(II) complex (10)>ligand (1)> Ni(II) complex (12)> Cu(II) complex (2)= Cu(II) complex (6)> Cu(II) complex (3)= Cu(II) complex (4) > Zn(II) complex (8)= Mn(II) complex (14). However, invivo studies for above compounds showed no toxicity was observed for all organs tested.
