Qualification of Metabolites in the Conditioned Medium of MSCs Cultured Using FBS and Platelet Lysate-A Comparative Analysis

Abstract: The immense potential of mesenchymal stem cells (MSCs) is exploited in cell-based therapy to treat acute and chronic pathological conditions. The in vitro propagation of MSCs is an absolute prerequisite for clinical applications. The umbilical cord tissue (UCT), being an abundant source of MSCs, have higher potential for ex-vivo expansion. High risk of transmission of zoonotic infections occur while using Fetal Bovine Serum (FBS), hence use of platelet lysate, the xeno-free supplement has displayed typical immunophenotype, plasticity, immunomodulatory property and chromosomal stability as per guidelines laid down by International Society for Cellular Therapy (ISCT). Conditioned medium enriched in growth factors secreted by these MSCs can be potentially used for cell free therapy. The present study reports the characterization of the conditioned medium using NMR spectroscopy. Analysis of spectra of 1D-NOESY enables the identification of metabolites in the biological samples from the available database.
