Formulation and study of some rheological properties of thermogelling gels of poloxamer 407 (Lutrol F127®) and native starch glycerol (Ipomoea batatas)

Abstract:

Justification. The aim of this work is to propose formulations based on starch gels and poloxamer 407 (P407) in order to obtain preparations with a higher contact time at the application site.
Goal : To develop thermogelling gels based on native starches and poloxamer 407 (F 127).
Materials and Methods. Pre-formulation tests of gels composed on the one hand, of P407 at 20% and sweet potato starch starch at different concentrations (1% to 10%, w/w) and on the other hand, of P407 at 20% and starch glycerol (GA at 2.5g, 5g and 10g) were performed. These gels were characterized by macroscopic tests, microscopic tests (polarized OPTICA optical microscope) and physicochemical tests. The formulation having presented the best profile, was selected for the continuation of this work. Also, rheological studies were carried out on it, using a rotary viscometer (HAAKE).
Results. The gel based on P407/2.5g GA showed good homogeneity, good rheofluidizing and thixotropic properties and thermosensitivity with a gelling temperature (Tgel) equal to 17.3°C. The pH was 6.7 at 37°C. Furthermore, the median particle size was 1.69 µm at 37°C. This could presage a good cutaneous applicability.
Conclusion. At the end of this work, the results obtained seem encouraging. However, the absence of active principle in the formulations, limit somewhat, the interpretation of the results obtained. Also, more thorough evaluations in the presence of a PA, should be carried out to better appreciate their impact on the physicochemical characteristics of these gels.
