Assessment of In-vitro Antimicrobial, Antioxidant and anticancer activity of bioactive peptides of the Goat Colostrum.

Abstract: The main aim of the present research communication is to assess the in-vitro antimicrobial, antioxidant and anticancer activity of bioactive peptides of goat colostrum milk extract. Goat colostrum not only contains proteins, lipids, carbohydrates, but they also provide immunoglobulins and bioactive peptides which help in building the immunity and protect against the microbial infections, growth and reduces the oxidative stress. In the present study, bioactive peptides were isolated from the goat colostrum and further these were screened and identified using HPLC and MALDI-TOF/MS. Antimicrobial activity was determined using agar gel diffusion method whereas antioxidant assay was determined using DPPH and FRAP analysis. The results of the antibacterial analysis of goat colostrum reveals that it’s potent against both gram positive (Bacillus subtilis and Staphylococcus aureus) and gram negative (Escheresia coli, Klebsiella pneumonia) bacteria. Anti-fungal analysis of goat colostrum also showing maximum growth inhibitory activity against Candia albicans (22mm) and Aspergillus niger (25mm). DPPH antioxidant activity of goat colostrum showed potent radical scavenging activity when compared with the positive control, ascorbic acid. The IC50 values for goat colostrum and ascorbic acid are 381.197 and 394.56 µg/ml respectively. FRAP method also showed maximum antioxidant potential of 90.454 mg of Gallic acid equivalents/g. The brain shrimp larvicidal LC50 was 10 ppm. The degree of lethality was directly proportional to the concentration of the extract. Maximum mortalities (100%) were observed at a concentration of 100 ppm and above concentrations. The bioactive peptides extracted from goat colostrum milk extract have potent cytotoxic activity against COLO 320DM cells and COLO 205 cells. Based on the results, the brine shrimp lethality of goat colostrum metabolites was found to be concentration-dependent.
