Antioxidant and antiparkinsonian potential of leaves and seeds of Moringa concanensis and Sesbania grandiflora in 6-hydroxydopamine induced parkinsonism in rats

Abstract: In the present study, ethanolic leaf extract of Moringa concanensis (EEMC), ethanolic seed extract of Sesbania grandiflora (EESG) and their combined extract (MCSG - CE) prepared using 1:1 ratio of their ethanolic extracts had been studied for their antioxidant and protective effect against 6-Hydroxy dopamine (6-OHDA) induced parkinsonism in rats. Forty-eight female Wistar albino rats (150-200g) were divided into eight groups with six animals each. Oxidative stress leading to neuronal damage similar to parkinson’s disease (PD) was induced in all rats by intracerebro- ventricular administration of 6-OHDA (8 µg/ 2 µl/rat) a powerful neurotoxin using stereotaxic apparatus. The ethanolic extracts of leaves and seeds ( EEMC and EESG) were prepared using ethanol by maceration and ethanolic combined extract (MCSG-CE) was prepared by taking EEMC and EESG in 1:1 ratio.Then rats were treated as follows: Gp-I: Normal control-Tween 80; Gp-II: Disease control- 6-OHDA;Gp-III: Standard- levodopa (6mg/kg, p.o); Gp- IV: EEMC-I(100 mg/kg, p.o.); Gp- V: EEMC-II (200 mg/kg, p.o.); Gp -VI: EESG-I(100 mg/kg, p.o.); Gp- VII: EESG-II(200 mg/kg, p.o.); Gp-VIII: MCSG- CE (EEMC (100 mg/kg) +EESG (100 mg/kg, p.o.) Antioxidant potential of extracts was evaluated by assessing malondialdehyde (MDA), catalase (CAT), superoxide dismutase (SOD)and glutathione (GSH). Antiparkinsonian potential was evaluated by estimating mono amino oxidase- B, dopamine and histopathological analysis. Treatment with Levodopa, EEMC, EESG, MCSG found to significantly decreased the MAO-B levels which correlated well with the increased dopamine levels.(*p<0.0001 and **p<0.05 were considered statistically significant). Neurodegeneration was milder in the drug-treated groups compared to the disease control. The antioxidant status was improved in the drug-treated groups which correlate with decreased MDA levels and increased CAT, GSH and SOD levels compared to disease control. Therefore, the present study concluded that EEMC, EESG and MCSG-CE can play promising role in antiparkinsonian treatment.
