Development of capsule formulations containing freeze-dried aqueous extracts of Lannea microcarpa Engl. and K. Krause (Anacardiaceae).

Abstract: Lyophilized powders of decocts of Lannea microcarpa have scientifically proven their efficacy and safety to treatment of arterial hypertension. However, these powders are very hygroscopic, with poor physicomechanical properties that cannot be used directly in capsules. Objective was to develop a capsule dosage form containing Lannea microcarpa's lyophilized powders (active principle). A pre-formulation was realized to evaluate the physicochemical and pharmacotechnical parameters of the powders, mixtures and granules. Wet granulation, with corn starch, colloidal anhydrous silica, magnesium stearate and polyvinylpyrrolidone as excipients, resulted in the granules. Two types of size 1 capsules were manufactured (directly with powders and with granules) and tested according to the European Pharmacopoeia 10th ed. Hygroscopic and flow properties of the extracts had a tackiness rate of 1.36 ± 0.5 requiring the addition of excipients. Extract-excipient mixture and the granules had different physicochemical properties. Excipients have made it possible to improve the rheological characteristics of powders. FC2 and FD3 formulations had the best galenic and biogalenic characteristics according to recommendations of European Pharmacopoeia 10th ed. Average phenolic tracer levels were 0.3922 ± 0.0096 mg gallic acid equivalent per capsule (GAE / capsule) for direct fill capsules (FC2) and 0.3920 ± 0.0079 mg GAE / capsule for granules (FD3). Dissolution of the two capsules showed that the releases of the tracer were greater than 80% after 45 minutes and in the three different solutions studied. This study allowed the galenic formulation of capsules based on extracts of characteristic good quality and potentially affordable for patient’s management.
