HPTLC-MS based quantitative analysis of carpaine in dried leaves extract of Carica papaya L. and its ADME analysis using SwissADME in-silico tools

Carica papaya L. (C. papaya) is a well-known traditional Indian medicinal plant used to treat various ailments such as dengue fever, diabetes, etc. Due to lack of quality and safety evidence, the present study is aimed for quantitative evaluation of carpaine in dried leaves of C. papaya using HPTLC method and its ADME analysis. The dried coarse powder of C. papaya leaves was extracted and fractionated using hexane, chloroform, and methanol. Thereafter, carpaine was isolated and identified with percentage purity using TLC-MS and HPLC techniques. Further, the method was validated by the HPTLC method for quantitative estimation of carpaine in C. papaya using chloroform: methanol (70:30, v/v) solvent system. Further, ADME analysis of carpaine was performed through SwissADME online tool to evaluate pharmacokinetics, drug-likeness and medicinal chemistry friendliness. The experimental findings suggest that the compound laid at the retention factor (Rf) of 0.61 was identified as carpaine through MS analysis with 91.23% purity through HPLC analysis. In HPTLC densitometric analysis, carpaine showed a linear curve plotted against peak area and concentrations over the range 200–2000 ng per band with a 0.989 correlation coefficient. The percentage recovery of carpaine was found to be 98.764-102.965% and the content was found as 0.537% (w/w) of alkaloids rich fraction. Further ADME analysis of carpaine through SwissADME online tool reveals that carpaine have good lipophilicity, drug-likeness, GI absorption and bioavailability. The present study enlightens that carpaine is the major alkaloid compound in C. papaya leaf and can be a key factor for its quality and regulatory aspects.
