Formulation and In-Vitro Evaluation of Mouth Dissolving Film Containing Drug Amlodipine Besylate

Abstract: The mouth dissolving films is most advanced form in the oral drug delivery system due to its flexibility and comfort which improves the efficacy of active pharmaceutical ingredients (APIs). The Amlodipine Besylate is calcium channel blocker used for treatment of hyper tension. The main objective of this work was to develop a mouth dissolving film of Amlodipine Besylate, allowing rapid dissolution in oral cavity, which improves bioavailability due to bypass of gastrointestinal tract (GIT) and bypassing first pass metabolism. The mouth dissolving film was prepared by solvent casting method with hydroxypropyl methylcellulose (HPMC) E15 as film forming polymer, glycerol as plasticizer, cross povidone as super disintegrants, citric acid as saliva stimulating agent and mannitol as sweetening agent. The films were studied for the various evaluation parameters. The physiochemical properties like general appearance of films, weight variation, thickness of films, surface pH , disintegration time , percent drug content , in-vitro dissolution study was evaluated which complies the specifications. Among all the formulated batches, formulation F8 shows best result which consist minimum concentration of HPMC E15 having drug release of 99.305 % at the end of 10 minutes. The drug release was best fitted to first order release kinetics and concluded that release profile depend on concentration of polymers.
