Impact of Maternal Hypothyroidism on Pituitary-Ovarian –Axis Activity in Females Offspring at Puberty

Abstract: This study designed to investigate the effect of experimentally induced maternal hypothyroidism during gestation and lactation on ovarian activity in both dams and their female offspring at puberty. Forty-eight pregnant dams were used divided into three treatment groups (n=8 each group) administered PTU 0.2 mg/kg B.W during gestation(GHpo), lactation(LHypo), and along the period of gestation, and lactation(G+L) Hypo, and three corresponding control groups (C1, C2, C3). Six female offspring born from each of the three groups of treatments (GHypoF, LHypoF & (G+L) HypoF) and a control group left untreated until puberty (60 after birth). Blood samples were collected from dams at the end of each experimental period and from female offspring groups at puberty. The results indicated a significant elevation in serum TSH and a significant reduction in serum T3 and T4 concentration were recorded in all PTU-treated dams groups compared with the controls. Nonsignificant elevation in serum TSH and reduction in serum T4 concentrations were recorded in both GHypoF & LHypoF groups compared with the control group while a significant elevation in serum TSH and reduction in serum T4 were recorded in the (G+L) HypoF group at puberty. Attenuated decrease in serum FSH concentration was recorded in LHypoF & (G+L) Hypof groups while a significant decrease was recorded in the GHypoF group compared with the control group at puberty. A reduction in antral follicles & corpus luteum and an increase in atretic and encysted follicles was observed.
