Evaluation of breast anticancer potential of geranyl caproate

Abstract:

Purpose: To increased anticancer activity of geraniol, geranyl caproate (GC) ester compound was synthesized.
The aim of this study was to evaluate anticancer potential of GC for human breast cancer.
Methodology: Synthesis of GC was carried out by batch esterificationn on the free solvent system. The reaction
was performed on the mol equivalent ratio between geraniol and caproic acid 1:1.2 in the present of 5%
NaOH catalyst. The effect of temperature (RT, 800and 1200C) and time (8 and 24 hours) on the reaction has
been studied. Serial techniques of spectroscopic methods namely FTIR, GC-MS, and NMR were used to
identify GC ester. Its potential as anticancer agent was in vitro analyzed through alamar blue method on human
breast cancer (MCF-7) cells and Mosmann method against normal (Vero) cells.
Results: Reaction at 800C for 8 hours produced highest yield of GC of 76.72%. It was found that GC was
potentially as breast anticancer compound indicated by IC50 value of 2.15 µg/ml against MCF-7 cells and very
less cytotoxic effect on Vero cells with IC50 value of 116.61 µg/ml. This gives an expectation that GC should be
used for breast cancer treating.
