Antioxidant, anti-tyrosinase, anti-elastase and cytotoxicity of ethanol extract from Cnidoscolus chayamansa (Mc Vaugh) leaf extract

Abstract: Cnidoscolus chayamansa (Mc Vaugh) is commonly name as “Chaya” or “Mexican spinach”, which is high nutritive value food and medicinal plant and use to treat or relieve several diseases by possessed biological activities. There is become favor to cultivate in Thailand and useful in Thai cuisine for noodle soup instead of other seasoning. Aims of this study were to evaluate antioxidant activities of ethanol extract from chaya leaf by DPPH inhibition, lipid peroxidation inhibition and nitric oxide radical scavenging assays; and to evaluate anti-tyrosinase, anti-elastase and cytotoxicity of chaya leaf extract. Fresh leaves were cleaned, air dried, and extracted with 95% ethanol by maceration. Total phenolic content (TPC) and total flavonoid measurements of chaya leaf extract was determined. DPPH, NO radical scavenging and lipid peroxidation inhibition activity of extract was evaluated and compared with ascorbic acid and ?-tocopherol, respectively. TPC contained in ethanol extract of chaya leaves was 14.32±0.59 mg GAE/g, however total flavonoid content was undetected. DPPH and NO radical scavenging activities of chaya leaf extract were poor when compared with ascorbic acid. Therefore, chaya leaf extract was possessed lipid peroxidation inhibition (IC50 = 129.27±7.78 mg/ml) when compared with a-tocopherol. Ethanol extract of chaya leaves was undetermined on anti-tyrosinase and anti-elastase activities. There was also no cytotoxicity of extract against human dermal fibroblasts. The results may useful data on different of antioxidant assays and other evaluation of biological activities from chaya leaf extract. Non-toxicity of chaya leaf extract was also confirmed in cell culture level.
