Isolated of calanon and 2.8-dihydroxy xanhone from Calophyllum sclerophyllum Vesque and their antioxidant and toxicity activities

Abstract:

In the present study, we are reporting the isolation, characterization, and bioactivity of Calophyllum sclerophyllum Vesque. The known calanon and 2.8-dihydroxy xanthone were isolated from the bark of C. sclerophyllum Vesque by using a combination of column and TLC (thin layer chromatographic) methods. Characterization of the pure isolated compound was performed by using LC-MS, FTIR, UV-Vis, and NMR analysis and comparison of their spectroscopic data white those reported in the literature. The toxicity and antioxidant activity of n-hexane, ethyl acetate (EtOAc), and methanol (MeOH) extract of the bark of C. sclerophyllum Vesque were done. Toxicity test was carried out by Brine Shrimp Lethality Test (BSLT), while antioxidant activity assay was investigated using the DPPH method. The results of the phytochemical screening showed that n-hexane fraction contained alkaloid and terpenoid, while EtOAc and MeOH fraction contains an alkaloid, terpenoid, saponin, tannin, and flavonoid. BSLT results showed that LC50 of n-hexane, EtOAc, and MeOH extracts were 197,72; 29,11, and 83,97 µg/mL respectively, while LC50 value of pure isolated compound (F6) as calanon was 316.89 µg/mL categorized as non-toxic. The EtOAc and MeOH extracts of C. sclerophyllum Vesque possess very active antioxidant activity with the IC50 value of 35.52 and 24.53 µg/mL respectively, while n-hexane extract and isolated compound F6 as calanon possess less antioxidant activity with an IC50 value 291.91 µg/m, 316.89 µg/mL respectively, and isolated F4 as 2.8-dihydroxy xanthone exhibited antioxidant activity with IC50 54.65 µg/mL
