Formulation and Development of Metoprolol Succinate Sustained-Release Matrix Tablet Using Eulophia Herbacea Tuber Mucilage as A Novel Polymer

Abstract: Background: Natural gums and mucilage’s are an alternative to synthetic drug release modifiers. These natural plant materials are cost effective, biocompatible, and biodegradable, and they have fewer side effects. The key purpose of this study was to formulate and characterize sustained-release tablets of Metoprolol succinate using Eulophia herbacea tuber mucilage as a novel polymer. Sustained release matrix tablets were prepared by wet granulation. The tablets were characterized for thickness, diameter, hardness, drug content, weight variation, friability, swelling index and in-vitro drug release. Results: The tablets passed all the tests required. The dissolution profile of the tablets revealed that the release of the drug was sustained up to 22 hours compared with a reference tablet formulation, PROLOMET XI 100. The dissolution profile data were fitted to release kinetic models such as zero order kinetics, first order kinetics, the Higuchi model, and the Korsmeyer-Peppas model to study the pattern of release of the drug from each formulation. The prepared sustained release tablet formulation was compared with a marketed formulation (reference formulation) in a dissolution study. The factors f1 (difference factor) and f2 (similarity factor) were determined. Conclusions: As the concentration of the Eulophia herbacea mucilage decreased, the rate of drug release from the formulation decreased. The best formulation was one that contained 20% Eulophia herbacea mucilage. Its drug release profile was comparable with the reference formulation with an f1 value of 10.9% and an f2 value of 62.9%. The study of the release kinetics indicated that the tablet formulation exhibits an anomalous (nonFickian) diffusion pattern and the release of drug is governed by diffusion-controlled as well as swellingcontrolled mechanisms.
