Evaluation of Hepatoprotective activity of Ochna obtusata extract on Paracetamol induced Hepatotoxicity in Rats

Abstract: Ochna obtusata was examined for its Hepatoprotective activity against Paracetamol induced liver damage in Rats since it has been reported to have antioxidant activity but the potential Hepatoprotective activity is still unclear and not reported. The objective of the study is to confirm the Hepatoprotective activity of ethanolic extract of Ochna obtusata. It has been evidently reported that administration of Paracetamol leads to development of Oxidative stress which consequently leads to alteration of hepatospecific serum markers. Thirty six rats were selected for the study and equally divided into 6 groups where the first three groups served as normal, Negative and Positive controls respectively. Groups IV, V and VI were treated with EEOO extracts of 100, 200 and 400 mg/kg respectively. Liver functions were carefully assessed for ALP, ALT, AST, ?-GT, LDH and Bilirubin levels, and antioxidant markers SOD and CAT. Pretreatment with EEOO prior to administration of Paracetamol significantly (P<0.05) helped to maintain the serum levels near to normal. Elevated levels of serum AST, ALT, ALP, LDH, ?-GT and bilirubin with significant (P<0.05) fall in levels of the marker enzymes in the liver tissue confirmed the Hepatocelluar damage. This study was also supported by elevated levels of lipid peroxidation (LPO) and a decrease in enzymic and non-enzymic antioxidants activities significantly (P<0.05). Histopathological evaluation amplified the study to assess liver for inflammation, infiltration or necrosis.
