Analysis of Eucalyptus Oil in Pharmaceutical Dosage Form: A Useful Approach for the Development and Validation of RP-HPLC Method

Abstract:

Eucalyptus oil, an essential oil known to exhibit different pharmacological activities like analgesic, antioxidant,
anti-inflammatory and also useful in aromatherapy, attributed to the presence of 1, 8-cineole as principal
component. Accordingly, a reverse phase HPLC method was developed for the estimation of oil and oil loaded
pharmaceutical dosage form using 1, 8-cineole as reference standard. It consisted of column phenomex C18
(5µm, 250×4.6 mm), isocratic pump mode, UV detection at 200nm, mixture of acetonitrile: water 70:30 v/v as
mobile phase and a flow rate of 1mL/min. The method was validated as per ICH guidelines, showed linearity
within a concentration range of 20-100µg/mL with R2 value of 0.9998. LOD and LOQ values were calculated as
0.017µg/mL and 0.175µg/mL respectively. The RSD values for repeatability, intermediate and method precision
were found to be within the limits of 2%. The recovery ranged between 100 to 102.48 % and robustness was
indicated by negligible variations in RT and peak area with deliberate variations in pH, flow rate and
temperature. The findings indicated that the proposed method was proved to be specific, simple, fast, sensitive,
precise, accurate and robust. Hence, the method was successfully used for the estimation of Eucalyptus oil in
pharmaceutical dosage form.
