Preliminary Phytochemical Screening with Evaluation of Antioxidant Activity of Merabilis Jalapa Leaves Cultivated in Iraq

Abstract: The aim of this study was investigation for the main active constituents in Mirabilis Jalapa plant, family Nyctaginaceae, widely distributed in Iraq. We extracted the leaves with different solvents increasing in their polarity. First we macerated the leaves in petroleum ether to extract non polar components, then we used dichloromethane to separate more polar compounds, later on we used acetone and finally methanol for separation of highly polar compounds. Then we concentrated all fractions for identification of the main active constituents with evaluation of antioxidant activity of each one. phytochemical investigation of Merabilis jalapa extract rich with many bioactive components like alkaloids, flavonoids, phenols, glycosides, tannins, saponins and lignin. Different fractions of leaf extracts were analyzed for the main active constituents by using High performance thin layer chromatography (HPTLC) with number of standards compounds by comparing the Rf values of standards [std1.rutin, std2. Kampferol, std3. P.coumaric acid, std4. Scopoletin, std5. Chlorogenic acid, std6. Coumarin, std7. Caffeic acid] with the Rf values of the separated components in all fractions, rutin, caffeic acids, coumarin were detected in all fractions while kampferol was only detected in dichloromethane and acetone fractions. Chlorogenic acid presents only in dichloromethane while P-coumaric acid only detected in methanol fraction. Then evaluation of antioxidant activity, there is a good correlation between antioxidant activity and extraction systems content for both acetone and methanol fractions.This research showed that Mirabilis Jalapa could be a potential natural source of new phytochemicals with high antioxidant activity.
