Development and Validation of Absorbance Subtraction Method for Simultaneous Determination of Atorvastatin Calcium and Sildenafil Using UV Spectrophotometer

Abstract: The objective of current research work was to develop and validate a simple, accurate, precise Spectrophotometric method for simultaneous determination of Atorvastatin Calcium and Sildenafil in pure form. Absorbance Subtraction technique has b??n used in simultaneous determination of both drugs by using UV Spectrophotometer. Methanol was used as solvent .The absorption maxima of Sildenafil and Atorvastatin Calcium was observed at 292 nm and 246 nm respectively. The Isobestic point was found to be 272 nm. Validation by UV Spectroscopy Method linearity range was determined by standard calibration method in the concentration range of Atorvastatin Calcium (3µg/ml to 27µg/ml) & Sildenafil (5µg/ml to 45µg/ml). The linearity study showed that the developed methods were linear at the specified concentration. LOQ for Sildenafil& for Atorvastatin Calcium were 0.046µg/ml and 0.044µg/ml and similarly LOD for both Sildenafil& for Atorvastatin Calcium was found to be 0.015µg/ml and 0.014µg/ml. Robustness & Ruggedness studies proved that the developed methods were found to be robust & rugged on deliberate changes done. A simple, accurate and precise UV-Visible Spectrophotometric method for the estimation of Sildenafil and Atorvastatin Calcium was developed and validated.
