Optimization of Xanthan Gum Production and Ethanolic Extract of Psidium Guajava Application in Dentistry

Xanthomonas campestris is widely known for its production of xanthan gum. Hence, in our study, we used shrimp shell (SSE), lemon peel (LPE), and yeast malt extract (YME) medium containing glucose as a carbon source to produce xanthan gum using the isolated strain of X.campestris. Our study compared the yield of xanthan gum using the aforesaid substrates and we found that YME medium containing glucose has given the maximum yield when compared with other substrates like shrimp shell, and lemon peel extract. Further, we optimized the yield of xanthan gum and found that the maximum yield was obtained in 10 d of fermentation in medium with LPE (0.30g) for 2% of LPE at pH-6.0. Further, we solubilize the crude bioactive components using ethanol and toluene from guava leaf powder. The bactericidal properties towards the isolated and characterized dental caries such as Streptococcus mutans and Lactobacillus acidophilus were studied using the crude ethanol and toluene extracts of guava leaf. The crude ethanol extract of guava leaves showed an excellent bactericidal activity than toluene extract as compared with reference positive control of chloramphenicol. Furthermore, we analyzed the crude ethanol extracts of guava leaf by GC-MS and we found that crude extract contains a wide variety of bioactive components. Hence, it can be manipulated as a toothpaste formulation and effectively used to preserve healthy teeth and protect against dental caries infection respectively. Interestingly, the xanthan gum obtained from the X.campestris also effectively used as a thicker in the anti-caries for the preparation of herbal toothpaste production in the future.
