Investigation of antioxidant potential of Lobophora variegata

Marine algae have received special attention as a source of natural antioxidants. Lobophora variegata is a species of small thalloid brown algae distributed mainly in the Gulf of Mannar. This study evaluates the antioxidant activity of L.variegata using nitric oxide radical scavenging and hydroxyl radical scavenging assay. The extracts of L.variegata exhibited good nitric oxide scavenging activity at low concentration thereby the concentration of nitrate in the assay medium gets diminished. As the concentration increases, there is a mild decrease in the activity. Among them, methanolic extract of L.variegata inhibited the accumulation of nitrite significantly, leading to the liberation of a stable oxidized product of nitric oxide from sodium nitroprusside (93.31%) at a concentration of 50µg/ml compared to the other three extracts [ Hexane (90.94%), chloroform (91.03%) and ethyl acetate (91.24%)]. When nitric oxide reacts with superoxide to form the peroxynitrite anion (ONOO-) its toxicity gets increases and the latter is also a strong oxidant that can decompose and produce OH (hydroxyl ion) and NO2 (nitrogen dioxide). The hydroxyl radical scavenging assay shows the abilities of the extracts (Hexane, chloroform, ethyl acetate, methanol) of L.variegata to inhibit hydroxyl radical-mediated deoxyribose degradation in a Fe3+ -EDTA-ascorbic acid and hydrogen peroxide reaction mixture. The results indicate that the methanolic extract showed the highest antioxidant activity with the inhibition of 99.02% at 50µg/ml. The results shown that methanolic extract produced significant antioxidant activity.
