Optimization and Evaluation of Developed Topical Ointment Formulation Containing Cefadroxil and Its Antibacterial Activity

Abstract: Skin disorders arise commonly and often contemporaneous in respect of therapeutic challenges to physicians due to the rising concerns on the topic of multidrug-resistant bacterial, viral, and fungal strains. Development of topical formulation against bacterial infection is still lacking and stealing economy. The present study is associated with the optimization and evaluation of developed tropical ointment formulation containing cefadroxil against bacterial infections. Twelve different ointments formulation were prepared and in-vitro drug release profiling was estimated to optimize developed formulations. Afterward, physicochemical, FT-IR and stability analyses were performed to determine the physicochemical compatibility of the optimized formulation. The antibacterial activity was performed against Staphylococcus aureus through zone inhibition assay. The resulted data revealed that the formulation C2 was selected as the optimized formulation which had the highest drug release affinity (94.14±0.45%) among all the developed formulation. Further, uniformity in drug content, pH, viscosity and spreadability of the optimized ointment formulation were found as 491.45±1.189 µg/mg, 5.987±0.443, 1476.0±6.867 pas, and 5.34±0.245 cm respectively. The FT-IR analysis of the developed formulation was showing all the principal absorption peaks of cefadroxil with no chemical interaction of formulation bases. Besides, stability analysis reveals no significant changes in pH, viscosity, spreadability and drug content of the optimized formulation. In antibacterial zone inhibition assay, the formulation C2 showed a strong zone of inhibition against the growth of Staphylococcus aureus. Hence the study confirms that the developed and optimized formulation can be a better pharmaceutical for bacterial infections and can be an alternative instead of economically expensive tropical formulations.
