Pulsatile drug delivery system – a novel approach for time and versatile rate-controlled drug delivery

Abstract: The area of pharmaceutical research is broadened with the invention of new pharmaceutical drug delivery system. The traditional drug deliveries systems have not the way to treat or cure special disease or as well as the common disease with less side effects and maximized efficacy. Oral drug delivery system (ODDS) has various advantages and disadvantages but their disadvantages are overcome by the controlled drug delivery system (CDDS). CDDS although they overcome the disadvantages of ODDS but do not handle the special pharmacological disease requirements. To overcome the disadvantages of CDDS thePulsatile drug delivery system (PDDS) is established. PDDS has gained importance in the drug delivery system because of improved patient compliance, therapeutic efficacy and having fewer side effects.Pulsatile mechanisms of drug distribution are configured to provide drugs in line with circadian disease activity. The substance follows a sigmoidal drug release profile marked by a time cycle without release and a fast and full release of the drug. The use of such technique helps medications to be administered in good time, in the proper dosage and at the right spot. Chronotherapeutics have researched and identified the possible effects for many illnesses including asthma, inflammation, cancer, diabetes, epilepsy, high blood pressure, ulcer, hypercholesterolemia, etc. Different unit systems modulated by soluble or erodable polymer coating are present on the market with capsules, osmotic membranes, single and multiple unit systems. These systems are useful for chronopharmacological conditions, where dosing is essential at night, medicinal products with strong first-pass effects, site-specific GIT absorption, or medicinal products with high toxicity and/or resistance danger. This systems also improve user conformity by reducing the dosing level.
