Cinchona ledgeriana is one of Kina plants species that have the highest levels of alkaloids. There are many extraction methods to obtained medicinal plant extracts. In this study, the extraction process for Kina leaves was carried out using soxhlet and maceration. This study aims to examine the effect of sonication pretreatment and extraction methods on yield and antioxidant activity. The sonication pretreatment process is carried out using two types of variables, namely time and amplitude. Antioxidant activity was measured using DPPH (2,2-diphenyl-1-picryl-hydrazyl) free radical scavenging method. Sonication pretreatment at 60 minutes and 90% amplitude gave highest yield of Kina leaves extract by soxhlet and maceration method respectively. The highest antioxidant activity was obtained from the soxhlet with an amplitude treatment of 90% for 60 minutes. In the maceration method, the highest antioxidant activity is a sample with a treatment of 90% amplitude and a time of 20 minutes.
