Comparative Anti-oxidant Efficacy of Polar and Nonpolar Fractions Obtained from Crude methanolic root extract of Asclepias curassavica (scarlet milkweed)

Abstract: Asclepias curassavica (Apocynaceae) plant has high medicinal value. The root extract, in particular are used as abortifacient, antihemorrhagic and as an expectorant for pnemonia. The free radical scavenging activity of A.curassavica leaf, flower, and latex extracts are reported in previous studies. But, the activity of roots has not been comprehensively studied so far. Hence, the study was designed with two objectives; first, to determine the in-vitro potential of methanolic root extract of A.curassavica as a natural antioxidant. Second, to comparatively assess the free radical scavenging efficacy of polar (methanol fraction) and non-polar (n-hexane fraction) phyto-chemicals present in A.curassavica roots. The crude extract effectively inhibited free radicals in DPPH assay (IC50=36.43±7.8), FRAP assay (1.13 µmol/ml FeSO4.7H2O), hydroxyl radical scavenging assay (IC50= 39.9 ±4.73) and anti-lipid peroxidation assay (IC50= 54.95 ±2.77). But, it was found to be less effective in scavenging superoxide anion radicals (IC50= 579.64 ±4.73), and nitric oxide radicals (IC50= 451.53 ±27.65) when compared with controls used in the tests. However, a concentration-dependent increase in free radical scavenging activity was observed in all tests. The total phenol content and total flavonoid content of the crude extract were 26.32±0.08 mg GAE/g of dry root and 18.1±0.04 mg QE/g of dry root respectively. The methanol fraction contained more phenols and flavonoids than n-hexane fraction. In all in vitro tests, the crude extract showed the highest activity, followed by methanol (polar) fraction. Least activities were recorded in n-hexane (non-polar) fraction. The study concludes that polar phytochemicals present in A.curassavica roots play a key role in the oxidative processes than non-polar constituents and A.curassavica root can be used as a potent antioxidant.
