In-Vitro Insulin Secretion and Glucose Uptake Assay of Different Extracts of Tinospora Cordifolia with their FTIR and GCMS Characterization

Abstract: In India, Diabetes has become a major metabolic disorder and about 40% population effected. Moreover, several drugs are being precedent for diabetes management in various systems of medicine in the country, but scientists are still in search of safe, effective and curable therapy. In the series of investigation, a study was carried out on various extracts of Tinospora cordifolia stem. The extracts prepared in water, alcoholic, 50% hydro-alcoholic solvents were evaluated for in-vitro models of insulin release in mouse insulinoma pancreatic cell lines (MIN6) and glucose uptake assay by using rat skeletal muscle cell lines (L6). None of the extract extract showed cyto-toxicity on both the cell lines evaluated in dose range of 1 to 1000 µg/mL. The selected fixed dose of extracts stimulated the insulin secretion in MIN6 cells model under 5 and 15mM concentration of glucose and increase glucose uptake in L6 cells. In comparison with standard glibenclamide, insulin and metformin, extracts were found to significantly release insulin and increase glucose uptake (P<0.005). All the extracts were also subjected to phyto-chemical investigations and found positive for alkaloids, flavonoids, tannins, glycoside and steroids. Screening of phytochemicals performed by using GCMS and FTIR techniques in which major compounds campesterol, stigmasterol, beta sitosterol, D-mannoheptulose, alpha amyrin, lupeol, alpha santonin, alpha carotin, columbin, glycine, betulinaldehyde and betulin were found in GCMS analysis.
