Antihypertensive Activity of Polyherboceutical Formulation in Different Composition and Each Ingredients on Albino Wistar Rats

Abstract:

The present investigation was carried out to evaluate the effect on blood pressure of aqueous extracts of root of
Rauwolfia serpentina (Rs), bark of Terminalia arjuna (Ta), root of Boerhaavia diffusa (Bd), whole plant of Convolvulus
pluricaulis (Cp), whole plant of Centella asiatica (Ca), rhizome of Nardostachys jatamansi (Nj) and polyherboceutical
formulations in normotensive, DOCA induced hypertensive rats and the anti-anxiety effect of formulations by light
and dark model on albino wistar rats. Acute toxicity of polyherboceutical formulation (PF) 01, 02 and 03 were
performed by oral route administration. This study revealed no acute toxicity was observed in all formulations,
hence all formulation were safe for use. PF 03 in 45mg/kg (by oral route) dose was studied in normotensive,
DOCA induced hypertensive rats using the non-invasive technique and in dark and light model (Antianxiety model)
with comparison of individual herbal extracts, polyherboceutical formulation 01 (27 mg/kg), polyherboceutical
formulation 02 (36 mg/kg) and standard drug as Verapamil (9 mg/kg), Diazepam (1 mg/kg) respectively. The results
showed that the polyherboceutical formulation 03 significantly (p < 0.001) decreased both systolic blood pressure
and diastolic blood pressure in DOCA induced hypertensive rats. Extract of Rauwolfia serpentina (Rs) and PF 03
significantly (p<0.001) increased the number of crossing (tunnel) and time spent in light and dark area when
compared to control group and standard group (Diazepam) which indicated the anti-anxiety activity. The results
indicate that PF 03 possessed antihypertensive and anxiolytic activity.
