Citrus flavonoids - nature’s boon in cancer therapeutics

Abstract: Flavonoids are richly found in fruits and vegetables, utilized as chemopreventive agents. The molecular study of flavonoids shows that they arrest the cell cycle, inhibits heat-shock protein, inhibits tyrosine kinase, decreased p53 protein expression, estrogen receptor-binding capacity, Ras protein inhibition. Proliferation of cancer cells has been inhibited by citrus flavonoids such as diosmin, rutin, apigenin, tangeretinin in different classes of human cancer cell line cultures. Flavonoids play a crucial role amongst the known inhibitors of angiogenesis. The major functions of citrus flavonoids are they act as reducing agents, metal chelators, scavengers of reactive oxygen species, antioxidants, singlet oxygen formation quenchers, and protectors of ascorbic acid. Flavonoids having that capability to inhibit DNA damaging, mutagenic signalling, cell proliferation and also proto-oncogenes (cFOS, cJUN, and cMyc). A systematic mechanism is to be developed to understand the flavonoids' mechanism when entering multiple cell organelles and tissues. Flavonoids are probable to inflect various biological actions in cancer, including apoptosis, vascularization, cell differentiation, and cell proliferation. The evaluation of pharmacokinetic parameters and the distribution of citrus flavonoids across biological fluids is essential. Future research on citrus flavonoids should focus on evolving a bio-system or method for exactly evaluating bio-accessibility and bioavailability in-vivo. To prevent or treat human cancers, long-term clinical trials should be performed to study the metabolism and cytotoxicity.
