Effects of Various Concentrations of Lysate Platelet-Rich Fibrin on Human Dental Pulp Stromal Cell Migration Activity

Abstract: Introduction: Pulpal healing is a complex molecular and cellular process; unlike other connective tissues, the received wisdom is that pulp has limited reparative ability. The development of a platelet-based secretome for use as a condition media (CM) has created the potential to influence the migration, proliferation, and differentiation of mesenchymal stromal cells (MSCs). However, no studies have examined the effectiveness of platelet-rich fibrin lysate (PRF-L) in induces migration of DP-SCs. Objective: This study analyzed the effect of various concentrations of a PRF-L CM on the migration activity (migration rate and relative wound closure) of human dental pulp stromal cells (DP-SCs). Material and Methods: Human DP-SCs were divided into six groups: Group I (control group): DP-SCs in 10% FBS CM; Group II: DP-SCs in 1% PRF-L CM; Group III: DP-SCs in 5% PRF-L CM; Group IV: DP-SCs in 10% PRF-L CM; Group V: DP-SCs in 20% PRF-L CM; Group VI: DP-SCs in 25% PRF-L CM. Migration activity in all groups was measured using a scratch wound assay after 24 hours. The cell characteristics were evaluated using a microscope to confirm the qualitative data. All statistical analyses were carried out using one-way ANOVA and a post hoc Bonferroni correction (p < 0.05). Results: There were significant DP-SCs migration activity differences amongst all the PRF-L CM groups compared to the control group (FBS CM). Conclusion: Various concentrations of PRF-L have different effects on the migration activity of DP-SCs. Compared to all the groups, the fastest DP-SCs migration activity occurred in the 5% PRF-L after 24 hours.
