Abstract: This study aimed to determine the effect of using Al2O3 active neutral in the process of andrographolide isolation from Andrographis paniculata. The extraction of Andrographis paniculata herbs was carried out with ethanol solvent. Isolation of andrographolide was continued using non-polar solvent, then passed to Al2O3 column and eluted with methanol. Next, the purification of isolated crystal was obtained by recrystallizing the extract using cold methanol. Identification of the isolated crystal was confirmed qualitatively by an analysis of the melting point, TLC, and FTIR spectra. The data obtained then compared with an analytical grade of andrographolide as the standard. The results showed that the data of isolated crystal was similar to the andrographolide standard from the parameter of melting point, TLC analysis, and FTIR spectra. From this research is known that isolation of andrographolide using Al2O3 could be an alternative method which more efficient and less solvent. In conclusion, this method is potentially developed as one of the methods to isolate andrographolide compounds from Andrographis paniculata
