Neuroprotective Potential of Efavirenz Nanoparticles on Cerebral Ischemic Stroke by Middle Cerebral Artery Occlusion in Wistar Rats

Present study aims to evaluate the neuroprotective repurposing potential of efavirenz by using in-silico, ex-vivo and in-vivo methods. Antioxidant activity was evaluated by DPPH and nitric oxide scavenging methods. The in-vivo neuroprotective effect of nanoparticles containing Efavirenz was carried out in cerebral ischemia induced through middle cerebral artery occlusion (MCAO) in rats. Behavioral alteration was measured by using Y maze and open field habituation. Biochemical parameters such as acetyl cholinesterase (AchE), Glutamate, Lactate dehydrogenase (LDH), Superoxide dismutase (SOD), Glutathione peroxidase (GPx) and catalase were estimated. Molecular docking studies suggested that efavirenz inhibits neurotoxicity inducing NMDA receptors activation, free radical scavenging potential of efavirenz was revealed by the comparison of ascorbic acid and in-vivo results envisage efavirenz on behavioral parameters improved in Y Maze and Open field indicates anti amnesic property of efavirenz. The inhibitory effect on neuron toxicity and improved levels of antioxidant enzymes contributes the neuroprotective potential of efavirenz but repurposing potential of efavirenz need further conformation to identify molecular mechanisms involved in neuron protection in cerebral ischemia.
