Effectiveness of Binahong Leaf Extract (Andrea cordifola) and Vitamine-E as a Candidate for Antiinflammatory Herbs in Acute Metritis Rat (Rattus norvegicus) to IL-6 Levels by Flowcytometry Method and Vaginal Histopathology

Abstract: Metritis is one of reproduction disorders which affects reproductive performance especially uterine involution with 20-40% incidency. Binahong leaf and vitamin E are used for their active compound as anti-inflammatory and antioxidant candidates. The objectives of this research are to determine the effectivity of binahong leaf extract and vitamin E on inflammatory cell infiltration feature in the histopathology of vagina and on the Interleukin-6 level. This research is a laboratory experimental research utilizing Complete Randomized Design (CRD). The measurement of IL-6 level used flowcytometry and the calculation of the number of infiltrating inflammatory cells was by using vagina histopathology preparates. Animal models used 18 female rats separated into 3 groups, which were positive control, negative control, and treatment groups. All female rats were induced by Staphylococcus aureus. Negative control group was without therapy, positive control group was given amoxycillin 1 mL therapy, while treatment group was given binahong extract 0,3 mL and vitamin-E 1 mL. Data analysis used SPSS One Way ANOVA with 95% level of confidence, followed by Least Significant Difference (LSD) test. The result showed that binahong leaf extract and vitamin-E in a dosage of 50 mg/ 200gram BW and 0,04 IU/animal can suppress IL-6 level lower compared to negative control group. Vagina histopathology observation showed that treatment was able to lower inflammatory cell infiltration compared to negative control with score 2. The conclusion obtained from this research is that binahong leaf extract and vitamin-E can be used as alternative herbal treatment for acute metritis.
