Formulation, Characterization and Evaluation of Ketoconazole Occuserts of ?-Cyclodextrin Complex

Abstract:

This research work aimed to formulate, characterize, and evaluate ketoconazole complexed with ß-cyclodextrin ocuserts for ocular delivery. The method of preparation involves three steps which involve ketoconazole reservoir film preparation followed by the addition of ethyl cellulose polymer as a rate controlling film and sealing the ocuserts. The formulated ocuserts were characterized and evaluated for FTIR, DSC, physical characteristics, film thickness, drug content, weight uniformity, pH, folding endurance, moisture absorption and its loss, swelling index, dissolution and stability studies. FTIR and DSC revealed to be no interactions with each other. The developed ocusert F 5 was white with a smooth texture, 0.17 mm of film thickness, 89.51% of drug content, 19.89 mg of weight uniformity, 91 folding endurance, 8.02 and 6.24 of moisture absorption and loss, swelling index of 13.7 mg in 60 hrs and drug release of 95.74 % for 20 hrs. The optimized formulation (F 5) was found to be stable upto 30 days. Therefore, we can conclude that prepared ketoconazole complexed ß-cyclodextrin ocuserts was found to be effective for ocular delivery.
