Evaluation of Antioxidant and Anti-inflammatory activities of Periploca aphylla

Abstract:
|
Aims: Phytochemical screening and evaluation of the antioxidant, anti-inflammatory and anti-microbial activity of Periploca aphylla Decne. locally called ‘Handabub’ to corroborate its ethno-medicinal use in treating inflammation, fever, cough and as a laxative.
Methods and Material: Following standard protocols phytochemical analysis was performed on methanolic extract of the aerial parts of P. aphylla. Anti-oxidant efficacy was evaluated by its DPPH radical scavenging potential and effectiveness in scavenging hydrogen peroxide radical. Quantitative estimation of the total phenols was done using Folin-Ciocalteu reagent assay, flavonoids were estimated using Aluminium chloride colorimetric method. In vitro anti-inflammatory activity was quantified by protein denaturation and proteinase inhibition. Relative anti-bacterial activity was checked using Disk diffusion method and Minimum inhibitory concentration (MIC) was evaluated using different dilutions of the extract.
Results: Phytochemical screening of Periploca aphylla, demonstrated the presence of flavonoids, glycosides, terpenoids, steroids, alkaloids, saponins, tannins, quinones and coumarins, while anthraquinones were absent. Extracts displayed substantial antioxidant potential, with an IC50 value of 68.33 µg/ml for DPPH radical scavenging and a strong hydrogen peroxide scavenging activity with an IC50 value of 113.82 µg/ml. Total antioxidant capacity was observed to be 244?±?2.7 mg AAE/ g. The total phenolic content and the total flavonoid content detected was 186±2.4 mg GAE/?g and 71. 466 mg of QE/g of dry extract respectively. Substantial inhibition of protein denaturation exhibiting an IC50 value of 108.02 µg/ml and considerable anti-proteinase activity ( IC50 value of 186.62 µg/ml) was observed indicating antiinflamatory capabilities. Antibacterial activity against 2 of the four tested bacterial strains was observed.
Conclusions: The results lend scientific support to the established traditional use of Periploca aphylla as an important medicinal plant.
