Assessment of sheep ova viability and morphology vitrified using new cryoprotectant solutions

Abstract: Back ground: Cryopreservation method, equilibrium rapid freezing (vitrification), used to preserve oocytes. Vitrification is a common method for cryopreservation of gametes. Honey has been used in many cultures for its medicinal properties, it has been used as a medical product since the earliest times. It has antioxidant activity and biological effects. It has greatly attracted the attention of researchers for its biological properties, have antimicrobial and antioxidant capacities. The chemical composition and the variety of beneficial nutritional and health effects of honey. Coenzyme Q10, boosts cellular energy and may enhance the immune system. It is an antioxidant produced in the body. Objectives: The present study was aimed to improve rate of oocyte viability and morphology using new solutions for cryopreservation, and thawing. Materials and Methods: Cryopreservation of oocytes using vitrification method, involving v two steps, once using common solution and again using new solutions, then thawing oocytes after 2 months and assessment oocyte viability and morphology. Collection of oocytes from sheep ovaries sheep after sacrifice, then assessment morphology and viability of oocytes using trypan blue stane for 30 seconds. Using vitrification solution (DMSO, EG), and once again using new solution with different concentration of honey (0.5, and 1.0 mL), then the oocytes viable loaded on vitricare invented by Prof Dr. Muhammad-Baqir M-R. Fakhrildin and in liquid nitrogen for 2 months, after this, thawing the oocytes by new solution using ( 20µm CoQ10) , and assessment the viability of oocytes. the SMART medium that used in the present research. Results: Results of the present study appeared significant increment (P<0.05) in the percentages of oocytes viability using new solution with honey as compared to the cryoprotectant solution. Conclusion: From the results of the present study it was concluded that the honey enriched to the new solution improved percentage of oocytes viability and no side effects on oocyte morphology.
