Formulation and Evaluation of Polymeric Film Loaded With Ibuprofen for Buccal Delivery

Abstract:

Objective: This study aimed to formulate badam gum (BG) buccal film (BF) loaded with ibuprofen (IB) for buccal delivery and showed better drug release from designed BF because the crystalline form of IB was converted into amorphous form during film formation.
Methods: IB loaded BG BF was prepared by the solvent casting method.
Results: Optimized buccal films with sufficient mechanical strength were prepared from the gel containing 1.0 % w/w of BG in combination with 0.4 % w/w pluronic F 127, 5.5 % w/w PEG 400, 3.0 % w/w hydroxypropyl methylcellulose, 1.0 % w/w polyvinyl alcohol and 0.9% w/w IB. The films were characterized using texture analysis (TA), hot stage microscopy (HSM), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), x-ray powder diffraction (XRPD), and in vitro drug release. The film contains an optimum ratio of 5.5 % w/w of PEG 400 for empty films (without drug) and IB loaded films, showed optimum film flexibility confirmed by TA. The residual water content of the prepared films was found to be 5.8 % using TGA. XRPD study confirmed the presence of amorphous form of IB. In vitro drug release study revealed that the amorphous form of IB showed better release than the crystalline form of IB from the polymeric film at saliva pH.
Conclusion: In conclusion, this study revealed that the prepared polymeric films have the potential for buccal drug delivery and transport drugs across the ex vivo buccal membrane in subsequent studies.
