Antidiabetic and Antioxidant Activities of Nanoemulsion Containing Mixed Herbal Extract in Alloxan-Induced Diabetic Rats

Abstract: Nanoemulsion containing mixed herbal extract of Andrographis paniculata (Burm f.) Ness. and Phyllanthus niruri, L have been developed to overcome lower bioavailability of the extract. In this study, the antidiabetic and antioxidant activities were substantiated and potential acute toxicity induced by the synthesized nanoemulsion was evaluated in the rats. Hyperglycemic condition was induced by the administration of alloxan. DPPH assay method was conducted to assess the antioxidant activity of the synthesized nanoemulsions in vitro. An assessment and evaluation of potential acute toxicity by the nanoemulsion was conducted in the rats at a dose of up to 2000 mg/kg. The mortality rate, increment of body weight, and possible changes in behavior were evaluated. The nanoemulsion was found to preserve the antioxidant activity both in vitro and in vivo. This was substantiated by the reduction in the level of malondialdehyde level in diabetic animals. There has been a significant level of hypoglycemic activity following the administration of 200mg/kg of nanoemulsion compared to the control. No mortality was reported in any of the tested groups. The observation has suggested that the synthesized nanoemulsion containing mixed herbal extract hold hypoglycemic and antioxidant activities did not denote significant systemic toxicity in the tested diabetic model.
