Morphological and ITS Based Characterization of Some Important Fungi Isolated from Desert Soil under Harsh environment

Abstract: In this study, four fungal isolates (PSU1, PSU2, PSU3 and PSU4) were isolated from the desert soil (45 ºC) in Alkharj-Riyadh region of Saudi Arabia. The fungal isolates were characterised by microscopic (100X) and ITS based sequencing approach by PCR amplification method targeting 18s rRNA gene, conserved regions. The morphological data showed filamentous and conidial form of fungal signatures were obtained. Based on the sequencing results of the ITS regions of RNA genes of these isolates, isolated fungi; PSU1, PSU2, PSU3 and PSU4 belongs to Aspergillus ustus, Alternaria sp., Aspergillus parasiticus, Aspergillus sp. respectively with the identity of (95.7%), (99.27), (99.08%) and (97.37) respectively. The phylogenetic relationship of these isolates was also analysed by using neighbour-joining method at 1000x bootstraps through MEGA X program. The phylogeny results showed the nearest species; PSU1 (Aspergillus ustus; 90%), PSU2 (Alternaria sp.; 69%), PSU3 (Aspergillus parasiticus; 90%) and PSU4 (Aspergillus sp. 70%). The appearance of these types of fungal species is now much adapted to the soil and may be good sources of industrial uses. In this study, producing fungi from desert soil would be used as a good industrial source for a production of thermostable enzyme, especially in lignocellulosic biomass degradation.
