Multidose studies in rabbits for determining pharmacokinetic and pharmacodynamic interactions of gliclazide with nutraceutical

Abstract: World Health Organization has mentioned that one of the most prevailing health complications across the globe is diabetes and it is the major reason for deaths due to comorbidities. Once the person is affected by diabetes, it is said to be a lifelong treatment and hence patients look after supportive therapy in general. Nutraceuticals are one such supportive substances highly preferred by patients as well being suggested by physicians across the world. In this context, the present investigation is sought to study the interactions of nutraceutical on the pharmacokinetics and pharmacodynamics of gliclazide which is one of the most popular anti-diabetic drugs used in the treatment of diabetes. Eugenol has been selected in the study. Based on the positive results obtained with single dose study in out research lab, the present investigation is planned to study the influence of multidose studies of nutraceutical followed by gliclazide treatment. Streptozotocin was used for inducing diabetes in healthy rabbits. Four different groups, each with 6 rabbits, are taken in the study. Group I is normal control, Group II is diabetic control, Group III is treated with gliclazide alone and Group IV is multidose treated with eugenol and gliclazide. The results clearly indicated that there is an advantage in both pharmacokinetic and pharmacodynamic parameters with combination therapy of multidose eugenol followed by gliclazide in diabetic rabbits compared with the treatment of gliclazide alone.
