Effect of the polymeric matrix on the physicochemical properties of the Diclofenac sodium loaded fibers in Orodispersible Tablets

Abstract:

The present study aimed to develop fibers of Diclofenac sodium (DS) by electrospinning technique and
compressed into an orodispersible tablet by direct compression. The DS was compatible with the added
excipients confirmed by the FTIR and DSC studies. There are no peaks of crystalline Diclofenac sodium visible
in the XRD patterns of Diclofenac sodium loaded Eudragit L100 fibers which imply that the drug-loaded in the
fibers was no longer present as crystalline, but fully converted into an amorphous state. The 1H NMR
spectrum shows separate peaks for Diclofenac sodium and Eudragit L100 thereby indicating that there is
intramolecular interaction between Diclofenac sodium and Eudragit L100, suggesting good compatibility. All
formulations showed significant improvement in the rate of dissolution as compared to the crystalline active
pharmaceutical ingredient. The results showed that the developed Diclofenac sodium loaded orodispersible
tablet formulation was stable and can be potentially used in rheumatoid arthritis.
