In vivo antigenotoxic potential of sesame oil against cyclophosphamide induced genotoxicity and oxidative stress in mice

Abstract: This study investigated the protective effect of sesame oil against cyclophosphamide (CP) induced genotoxicity and oxidative stress in the murine test system. In vivo bone marrow chromosomal aberration, micronucleus and sperm abnormality assays were employed for the study. Hepatic antioxidant studies were also performed. Animals were divided into six groups of six animals in each. They were negative control (NC), positive control (PC-received CP 40 mg/kg) and three doses of sesame oil (1, 2 and 4 ml/kg) along with single dose of CP. Animals received sesame oil via oral route for two weeks while CP was administered intraperitoneally, 24 hours prior to sacrifice. The results demonstrated that different doses of sesame oil significantly reduced the frequencies of aberrant metaphases (AM) and chromosomal aberration (CA) as compared to CP treated PC animals. In addition, frequencies of micronucleated polychromatic erythrocytes (MNPCEs) and cytotoxicity were significantly suppressed against CP. Besides, sesame oil also protected the sperm cells against clastogenicity of CP as a significantly decreased percentage of abnormal sperms were observed as compare to PC animals. Hepatic antioxidant parameters revealed that sesame oil is capable to improve the glutathione (GSH), superoxide dismutase (SOD) activities and suppressed malondialdehyde (MDA) in the aganst CP. From present study it can be inferred that sesame oil is protective against CP induced genotoxicity and oxidative stress in mice.
