The report number of the adulterated herbal product by synthetic drugs have increased. This study proposed an analysis method to identify synthetic drugs in the herbal product to guarantee the herbal product's safety, quality, and efficacy. We employed Fourier Transform Infrared Spectroscopy Attenuated Total Reflectance (FTIR-ATR) coupled with multivariate analysis in this study to examine prednisone and diclofenac sodium in herbal products mainly Indonesian traditional herbal medicine called Jamu. The binary mixture of Jamu and synthetic drugs, Jamu samples, prednisone, and diclofenac sodium were recorded at specific range of wavenumber. Principal Component Analysis (PCA) and Discriminant Analysis (DA) visualized the classification group of samples, while Principal Component Regression (PCR) and Partial Least Square Regression (PLSR) were used for multivariate calibration. The score plot and Cooman's plot exhibited an ideal performance to discriminate the samples. The discriminant analysis presented an accuracy level >99,9% for grouping the samples. Multivariate calibration using PLSR and PCR gave an accurate and precise result according to the low value of RMSEC and RMSEP and R2 value almost 1 (>0,99).
