Long Term Intake Of High Protein Diet Reduces Body Weight And Glucagon Like Peptide-1 (Glp-1) Concentration In Male Wistar Rats

Abstract: Background High Protein Diets (HPD) have been widely used as a promosing strategy to achieve weight loss due to its double effects by increasing satiety and reducing fat mass. The high composition of dietary proteins found in HPDs are believed to stimulate the secretion of the gut hormones. The aim of this study is to determine the effects of an 8-week intake of high protein diet on Glucagon like peptide-1 (GLP-1) and ghrelin levels in male Wistar rats. Methods and Materials A total of 20 male Wistar rats were divided into two groups receiving either HPD (78.25% protein, 10.5% carbohydrate, 7.08% fat) or standard diet (15.25% protein, 52.2% carbohydrate, 7% fat). All rats received daily 30 g of their consecutive diet for 8 weeks. Blood samples were obtained at the end of the study to measure the GLP-1 and ghrelin levels using the ELISA technique. Wilcoxon, Mann-Whitney U, and unpaired t-test were used to analyze the data. Results HPD-fed rats lost a significant amount of body weight (154±13.1 vs. 268.2±8.5 gram, p<0.001) from week-2 through week-8 compared to the control group. In addition, HPD intake also significantly reduced the concentration of GLP-1 in the HPD-group rats (344.1±29.3 vs. 705.1±45.3 ng/ml, p<0.001) compared to the control group, but did not result in any difference of ghrelin levels between the two groups. Conclusion An 8-week intake of HPD significantly reduced body weight and GLP-1 levels in the HPD-group mice.
