Expression of Umami Taste Receptors on Rats (Rattus novergicus) at Different Levels of Food Hardness

Abstract: Affiliated Program for Children Development at University of George Town says, there are 6 types of eating difficulties in children, about 27.3% of children only want to eat liquid or crushed food and 24.1% of children have difficulty in sucking, chewing or swallowing. The hardness of food is important for determining one's appetite. The umami taste is the fifth sense of taste that is acknowledged as one of the basic tastes. Umami is considered a delicious taste that can arouse the appetite. The aims of this study to determine umami taste receptor TAS1R1 and TAS1R3 expression on rats (Rattus novergicus) that fed with different levels of food hardness. Post-weaned wistar rats (Ratttus novergicus) were divided into three groups, Group1 standard diet, group2 soft diet, group3 hard diet. The rats were sacrificed in 8th weeks and tongue was observed through scorring count TAS1R1 expression in fungiform and TAS1R3 expression in foliate with immunohistochemistry method using polyclonal antibody. The mean and standard deviation (X ¯ ± SD) of TAS1R1 expression on Group1 was 7.82 ± 1.66, Group2 was 7.37 ± 1.33, and Group3 was 4.84 ± 1.44. In group3 was significantly lower than group2 and group1. TAS1R3 expression on Group1 was 5.17 ± 2.58, Group2 was 8.55 ±1.85, and Group3 was 7.37± 2.32). In group2 was significantly higher than group1 and not significantly higher than group3. Hard diet group showed lower TAS1R1 expression, and soft diet group showed higher TAS1R3 expression.
