Increased Reproductive Capacity of Sprague Dawley Male Rats Assessed from the Number of Leydig Cells, Sertoli Cells, Primary Spermatocytes, and the Diameter of The Seminiferous Tubules through the Effect of Methanol Extract, Soluble and Insoluble Fractions Of N-Hexan Parijoto Fruit (Medinilla Speciosa Blume)

Abstract: One of the health problems in Indonesia is infertility, the incidence rate reaches 15-20% of about 50 million couples, where men play a role in about 50% of all cases. Parijoto fruit (Medinilla speciosa Blume) is empirically used by the people of the Kudus area, Tenah Java Province, Indonesia to increase fertility. The purpose of this study was to determine the potential of methanol extract, soluble and insoluble fraction of n-hexane parijoto fruit in increasing the reproductive capacity of male SD rats, studied from the number of leydig cells, Sertoli cells, primary spermatocytes, and seminiferous tubule diameter. The study was conducted with a posttest only control group design, using 24 male Sprague Dawley rats aged 2-3 months with 200-300 grams of weight divided into 6 groups. Groups I (normal), II (extract), III (soluble fraction of n-hexane) and IV (insoluble fraction of n-hexane) were each given a dose of 500mg/KgBW orally for 14 days. Observations were made on the 15th day and analyzed using Kruskal Wallis and continued by Mann-Whitney. The results showed that there were significant differences between each group. The mean seminiferous tubule diameter for groups I, II, III and IV were 260 ± 15.02; 322.9 ± 2.35; 291.55 ± 26.39 and 330.79 ± 9.06, while the mean number of primary spermatocytes was 26 ± 0.9; 32 ± 1; 28 ± 0.92 and 38 ± 3.11. The mean value of the number of Leydig cells 25 ± 0.65; 27 ± 1.00; 28 ± 0.75 and 31 ± 1.40 while the mean number of Sertoli cells was 27 ± 0.75; 32 ± 1.60; 29 ± 1.64 and 38 ± 3.36. The methanol extract, soluble fraction, and n-hexane insoluble fraction at a dose of 500mg/KgBW of fruit had a significant effect on the number of Leydig cells, Sertoli cells, primary spermatocytes and seminiferous tubule diameter.
