Phylogenetic characterization based on Mitochondrial cox 1 gene analysis, antibacterial and antioxidant potential of the red algae Hypnea cornuta extracts

Abstract: The study focuses on determining the antioxidant properties and the antibacterial activities of the different solvent extracts, namely methanol, ethyl acetate, and hexane, from the red algae Hypnea cornuta gathered from the Red Sea, Egypt. The nutrients of this alga have been studied by measuring total lipids, total soluble proteins, and total carbohydrates in its dry body weight. The algal sample was taxonomically identified using a recent molecular approach; phylogenetic analysis was based on cox1 gene comparison (BLAST) with sequences deposited in the NCBI gene bank. Data revealed that the gathered alga was Hypnea cornuta. The algal extracts showed high total antioxidant capacity (TAC) in the order of methanol ? ethyl acetate ? hexane extracts; (2.17, 1.87, and 1.53 mg equivalent ascorbic acid/g dry weight; respectively). The tested algal extracts showed also relatively high antibacterial potential against human and fish pathogenic bacteria; Gram-positive bacteria ((Staphylococcus aureus), and six Gram-negative bacteria (Salmonella spp., Vibrio angullarum, Aeromonas hydrophila, Pseudomonas aeruginosa, Escherichia coli, and Edwarsiella tarda). Interestingly, the algal tissues contained relatively high values of the nutrients such as total lipids, total proteins, and total carbohydrates, the matter which reflects the importance of this red alga as a resource full of nutrients and its promising medical applications as an antioxidant and antibacterial dietary food supplement.
