Formulation and Evaluation of Polymeric Orodispersible Tablets Contains Ibuprofen Loaded Microfiber for Better In-Vitro Dissolution

Abstract: Aim: The aim of the present study was to compress the Ibuprofen (IB) loaded polymeric microfiber Orodispersible tablets (PMOT) for better in vitro dissolution. Method: Ibuprofen (IB) loaded microfiber compressed into polymeric Orodispersible tablets by direct compression. Result and Discussion: 18% w/w Badam gum gel was used to prepare drug loaded micro fiber. Controlled sized polymeric microfibers were compressed into orodispersible tablets. Microscopic photographs showed that drug loaded micro fibers were clear, and transparent fibrous structure. More amount of drug was loaded in microfibers than physical mixture. Drug release profiles of both formulations carried out in distinct dissolution media. Double amount of drug Ibuprofen released at pH 6.8 from the micro fiber based tablets than directly compressed physical mixtures contains same composition. Polymeric chains of Badam Gum altered due to the process of fiber formation and micro-fiber based formulation showed better in vitro dissolution X-ray diffraction study revealed that drug loaded microfibers contains amorphous form of Ibuprofen whereas crystalline form was in the physical mixture. Microfibers contains citric acid enhances the in vitro dissolution of drug by changing pH of the dissolution media. Citric acid acts as a solid plasticizer by forming secondary bonds with the hydroxyl groups of Badam Gum altering the polymeric chains.
