Proliferation therapy, also known as “Prolotherapy,” is a regenerative injection therapy (RIT). Since the late 1930s, this technique has been used to stabilise injured joints and to relieve joint pain. This is now used in the treatment of Temporomandibular Disorders (TMD). This review aims to analyse the existing literature on the role of prolotherapy in TMD management. PUBMED and Google Scholar were searched to identify all clinical trials evaluating prolotherapy in the management of TMJ hypermobility, dislocation, and chronic pain of TMDs. The included studies were 4 clinical trials and one case report. From the included studies, prolotherapy was proven to be effective in patients with TMJ Hypermobility,Dislocation and chronic Pain with TMDs.All patients reported significant reduction in pain, clicking sound, locking episodes and associated symptoms. The quality of life of the patients showed good improvement. This review highlights the efficacy of prolotherapy in reducing the TMJ disorders in a majority of patients who have reached a plateau with use of an intraoral appliance, physical therapy, and home care. We recommend conducting more studies with larger sample size and with a sufficiently longer follow up period.
