Antimicrobial activity of quercetin from Majapahit (Crescentia cujete L.) fruit extracts: an in silico study

Abstract:

Aim: The aim of this research was to analyze the interactions of quercetin compound of Majapahit fruit fraction against Vibrio harveyi, a bacteria which caused vibriosis disease in shrimp, by in silico approach.
Materials and Methods: LC-MS was used to confirm the presence of quercetin, and the biological activity was predicted using PASS Online software. The mechanism of quercetin as an antimicrobial against Vibrio harveyi was investigated through the molecular docking of quercetin with DnK and gyrA receptors.
Results: The results of LC-MS analysis of Majapahit fruit (Crescentia cujete) fraction confirmed the presence of quercetin compounds. Quercetin has biological activities particularly have potential activity as an antimicrobial agent with Pa value (probability activity), including anti-protozoa (0.445), antiviral (0.484), antifungal (0.490), anti-tuberculosis (0.420), anti-mycobacterial (0.421), while the value of antibacterial was 0.393; if the probability activity (Pa> 0.3), the compounds computationally have potential as antimicrobial or antibacterial. The binding affinity for DnaK and gyrA was -9.0 kcal/ moll and -7.4 kcal/moll, respectively.
Conclusion: Majapahit fruit (Crescentia cujete L.) have potential as antimicrobial based on in silico approach.
