A Study on The Differential Gene Expression Pattern of Interleukin-6 Family Genes and Their Effect on The Survival of Hnscc Patients - An in-Silico Approach

Abstract: Head and neck squamous cell carcinoma (HNSCC) ranks among the top ten common forms of cancer prevalent world-wide. The incidence of HNSCC has shown a steep increase in recent years especially in the south and east Asian countries. The major risk factor identified to be associated with HNSCC are smoking, alcoholism, using pan, gutka etc., infections with human papilloma virus, poor oral hygiene, sharp tooth etc., Despite advancements in the diagnosis and treatment of HNSCC, the number of new cases identified on a day to day basis increased dramatically. The present scenario underscores the need for a more effective diagnostic method for early detection of HNSCC cases. In line with the above fact, the present study was designed to investigate the gene expression profile of IL6 members to derive a putative association between the disease phenotype. The study follows a retrospective, observational study design, which utilized the TCGA (The Cancer Gene Atlas) dataset hosted in UALCAN database. The gene expression profile for each of the eight genes of the IL6 family viz.,CLCF1, CNTF, CTF1, CTF2P, IL6, IL11, LIF and OSM was investigated. Among the eight genes analyzed in IL6 family, four gene showed significant difference in the expression pattern between the normal and the primary tumors. Furthermore, differential expression profile and survival curve analysis was also recorded for the four genes. The results of this In silico study opens new avenues towards identification of novel biomarkers which can be used for the prognosis and diagnosis of diseases. Further experimental validation is warranted to confirm the association of the four genes with HNSCC cases in the south Indian population.
